Believe it or not the best software for that is usually included with the sound card.
Provide us with some information and/or details about your sound card.

If you have Windows Vista or Windows 7 then there is a built in channel tester.

Under "Control Panel" go to "Hardware and Sound" and then "Sound". In the popup select "Speakers" and press the configure button. Under that select your setup (stereo, 4 channel, 5.1, or 7.1) and press the test button. It will go through each channel individually with a chime sound.
